RNA反义纯化技术(RNA Antisense Purification,RAP)运用于研究RNA与DNA、RNA及蛋白质之间相互作用的实验。根据研究对象不同,RAP技术结合了高通量测序(RAP-Seq)和质谱技术(RAP-MS)分别研究与目标RNA互作的基因和蛋白质。
RAP技术是通过设计生物素或链霉亲和素探针组,把目标RNA拉下来以后,与其共同作用的DNA、RNA或蛋白质(RBPs)就会附在到磁珠上,最后把基因做高通量测序,这样会得到该调控RNA转录调控的下游靶基因。与此同时,RBPs可以开展Western Blot验证或者MS鉴定未知蛋白。
