实验介绍

染色质免疫沉淀(ChIP)是研究体内蛋白质与DNA相互作用的理想技术,是目前研究转录因子(transcription, TF)与启动子(promoter)区域间相互联系的较好方法。ChIP与其他实验技术相结合,可用于不同用途的检测。ChIP-Seq方法特异性地富集与目的蛋白结合的DNA片段,并对富集得到的DNA片段进行高通量测序,从而确定该转录因子与基因组的结合区域。 (本试剂盒仅供科研用途)

实验原理:


在活细胞状态下使细胞内的蛋白质和DNA交联,并用超声波将其随机切断为一定长度范围内的染色质小片段。选择与目的蛋白特异性结合的抗体免疫沉淀DNA-蛋白质复合体,从而特异性地富集与目的蛋白结合的染色体片段。
