FISH探针
实验介绍

FISH探针是指能与特定核酸序列发生特异性互补杂交,杂交后又能被特殊方法检测的已知并被标记的核苷酸序列。FISH探针通过标记上荧光染料标记,后续用于可视化目标DNA序列的位置和数量。在确定目的基因序列后,选择与目的基因序列互补的序列片段作为探针序列,并根据实验条件选择合适的荧光标记物进行标记。

实验介绍:

FISH探针是指能与特定核酸序列发生特异性互补杂交,杂交后又能被特殊方法检测的已知并被标记的核苷酸序列。FISH探针通过标记上荧光染料标记,后续用于可视化目标DNA序列的位置和数量。在确定目的基因序列后,选择与目的基因序列互补的序列片段作为探针序列,并根据实验条件选择合适的荧光标记物进行标记。

探针类型:

①短链/寡核酸探针(15-40bp)是利用荧光染料标记到单链DNA上,通常使用化学合成方法,这可以通过磷酸化修饰或末端标记的方法将荧光染料引入到DNA分子的5' 末端或内部;

②长链探针(150-400bp)是将DNA分子的一端进行荧光染料标记,形成互补链5' 端带有荧光标记的DNA双链,通常使用PCR的方法进行标记将荧光染料引入到DNA分子其中一条链的5' 端。

标记类型:5' FAM/CY3/CY5等荧光标记。
